Everytime before i take my MBP out for a gig i use OnyX and clean out stuff, fix permissions and run all the regular clean up scripts. Than i power down and leave it powered off until im at the gig and EVERYTHING IS CONNECTED/POWERED ON than i power on my laptop.

This has worked everytime tried and true.

Honestly internal vs external does not "really" matter; internal will be FASTER because the SATA interface has more bandwidth and throughput than your USB/FW will. Also dont you want less crap to drag around to the club? so internal saves you plugins and extra USB crap which is always a pain to keep track of when the night goes on and the dj booth gets all rat nested lol.
